Immunomodulatory Properties of Wharton’s Jelly-Derived Mesenchymal Stem Cells from Three Anatomical Segments of Umbilical Cord

Mesenchymal stem cells (MSCs) are multipotent progenitor that reported to be immune-privileged and immune-evasive. MSCs capable of differentiating into specific cell types for subsequent use in cell-based therapy. They express low levels human leucocyte antigen (HLA)-ABC no HLA-DR. Wharton’s jelly-derived (WJ-MSCs) were also found leukocyte G (HLA-G), which renders them immunosuppressive. This study aimed determine whether cultured WJ-MSCs retain their immune-evasive properties after differentiation, these differ among derived from different anatomical segments the umbilical cord. Umbilical cords healthy pregnant mothers undergoing caesarean section obtained grouped by three segments: fetal, middle, maternal segments. isolated, culture-expanded, differentiated osteogenic cells. Expression HLA-DR, HLA-ABC, HLA-G quantified using flow cytometry. Both undifferentiated osteodifferentiated subsequently co-cultured with allogeneic peripheral blood mononuclear with/without lipopolysaccharide (LPS) stimulation five days. Lymphocyte proliferation assay was performed carboxyfluorescein succinimidyl ester (CFSE) as a tracker. Our results showed significant difference existed HLA profiles between without differentiation. Mean HLA-G, HLABC, HLA-DR 24.82±17.64, 52.50±18.41, 1.00±1.68%, respectively. Stimulation LPS increased blooc (PBMC) proliferation. However, PBMC significantly lower when PBMCs (p < .05; p .001 stimulation) than they WJ-MSCs. These findings suggest stimulate lymphocyte not immune-privileged.
